Gene TM1459 from Thermotoga Maritima encodes the protein NP_229258 whose structure (1vj2) is the first representative of a novel subfamily of cupins (PF07883) containing manganese that is coordinated by four histidines (His52, His54, His58, and His92) [Ref].
TM1459 has similar phylogenetic cooccurrence (string.embl.de) as TM0243 (a member of SAM radical superfamily).
1vj2 classifies inside the SCOP all beta class, RmlC-like cupins superfamily, TM1459 family. DALI top hits are with the RemF polyketide cyclase PDB:3ht1 (Z=17), a putative oxalate decarboxylase PDB:1o4t (Z=16), and the cupin-2 protein PDB:2pfw (Z=15). RemF also has significant (36%) sequence homology to TM1459 but shows a two-helix C-terminal extension of unknown function. Despite very similar coordination chemistry and geometry, the metal in RemF was identified as zinc or nickel, depending on the crystallization conditions [Ref].
Analysis of the crystallographic packing of 1vj2 using the PQS server {Henrick, 1998 #73} indicates that a dimer is the biologically relevant form. The N-terminal beta-strand is part of the beta-sheet of the opposite subunit, suggesting a bona fide dimer.
