Protein Summary
The TM0665 gene of
Thermotoga maritima encodes a pyridoxal 5'-phosphate dependent O-acetylserine sulfhydrylase, that catalyzes the beta-replacement of the acetyl group in O-acetyl-L-serine
by sulfide to generate L-cysteine with production of
acetate (EC 4.2.99.8; CysK; COG0031, PubMed:
15211522). This enzyme can also generate S-sulfo-L-cysteine using thiosulfate instead of sulfide. Metazoa and fungi does not have proteins homologous to TM0665, where cysteine is derived from methionine, but not from serine as in bacteria, archaea, and plants.
Protein encoded by TM065 has tryptophan synthase beta subunit-like PLP-dependent enzymes fold (SCOP sunid: 53685) with two similar domains related by pseudo dyad. Each domain has 3 layers (a/b/a) core with parallel beta-sheet of 4 strands (order 3214). Please note, PDB structure 1j6n of TM0665 was replaced by PDB structure 1o58.
