Protein Summary
The TM0140 gene of Thermotoga maritima encodes an indole-3-glycerol phosphate synthase (IGPS) (EC 4.1.1.48, PF00218), an enzyme catalyzing the fourth step in the biosynthesis of tryptophan. The 1j5t structure comprises a single-domain (beta/alpha) 8 barrel (TIM barrel). With the exceptional of an additional N-terminal helix (missing from TM0140), the structure is essentially identical (main chain rmsd 1.6 Å over 207 residues) with the IGPS of Sulfolobus solfataricus, another hyperthermorphile, for which the active site and catalytic mechanism have been described (Hennig 2002) (Fig. 1).

Figure 1. Ribbon diagram showing the superposition of the IGPS structures of T. maritima (PDB id: 1j5t, in blue) and S. solfataricus (PDB id: 1a53, in gray). The catalytically important residues are labelled for TM0140 and shown in ball-and-stick. The ligand (IGP) is shown in red.
